Cagrilintide

Native human amylin is notoriously difficult to work with: it aggregates into amyloid fibrils under ordinary buffer conditions, which makes reproducible in vitro work nearly impossible. Cagrilintide is engineered around that problem, borrowing stabilising residues from salmon calcitonin-family sequences and adding a lipidation handle for half-life extension. The result is an amylin receptor agonist that stays in solution long enough to study.

It is most often paired with a GLP-1 agonist in the literature because the two act through separable satiety pathways — amylin receptors in the area postrema, GLP-1 receptors more broadly — and the combination is used to probe whether the effects are additive. Lots are released with a supplementary aggregation check by dynamic light scattering.
